What they found
The trial targets not a disease but “ageing” as an endpoint: delaying the COMPOSITE first occurrence of cancer, heart disease, dementia and death — if it succeeds, the FDA would for the first time recognise ageing as a treatable target. Background: in cohorts, diabetics on metformin outlived non-diabetics (Bannister 2014).
⚠ Caveat
Not yet fully launched; newer data weaken the hypothesis: in the ITP metformin didn't extend mouse lifespan, the Keys 2022 analysis attributes the Bannister result to methodological bias, and metformin blunts exercise adaptation (Konopka 2019).
